HAV Hepatitis A Virus Ab IgG/IgM Test Cassette Operation Steps

1. Pre-Test Preparation

Ensure the test cassette is at room temperature (15–30°C). Gather required materials: test cassette, specimen collection device (e.g., sterile swab or dropper), disposable gloves, and a timer. Check the expiration date and integrity of the cassette package; do not use if damaged or expired.

2. Specimen Collection

For serum/plasma: Collect venous blood using a sterile needle and syringe, then transfer to a clean tube. Allow clotting for 10–20 minutes, centrifuge to separate serum, and use within 2 hours or store at 2–8°C for up to 48 hours.

For whole blood: Use fresh capillary blood from a fingertip puncture. Avoid hemolyzed or contaminated specimens.

3. Test Execution

Step 1: Open the Package

Remove the cassette from the foil pouch and place it on a flat, dry surface. Use immediately after opening.

Step 2: Add Specimen

Using the provided dropper, add 3 drops (≈100 µL) of serum/plasma or 4 drops (≈120 µL) of whole blood into the specimen well (labeled “S”). Take care not to cross-contaminate the dropper.

Step 3: Add Diluent (if required)

Some kits may require adding 1 drop of diluent (provided) after the specimen. Follow the manufacturer’s instructions carefully.

4. Incubation and Timing

Start the timer immediately after adding the specimen. Allow the cassette to react at room temperature without disturbance. Do not incubate at higher temperatures or expose to direct sunlight. Read results at 15–20 minutes. Do not interpret after 20 minutes, as prolonged incubation may cause false results.

5. Result Interpretation

Positive Result:

Two distinct colored lines appear: one in the control zone (C) and one in the test zone (T) for IgG or IgM, indicating past or current HAV infection.

Negative Result:

Only the control line (C) appears, indicating no detectable HAV antibodies.

Invalid Result:

No control line (C) appears, suggesting improper handling or a defective cassette. Repeat the test with a new cassette.

6. Post-Test Procedures

Dispose of used cassettes and specimens in biohazard waste containers according to local regulations. Clean and disinfect work surfaces. Record results in the patient’s medical file, noting the test type, date, and time.
